- This antibody is designed, produced, and validated as part of a collaboration between Rockland and the National Cancer Institute (NCI). Int6 is a candidate tumor suppressor in multiple neoplasms, and in particular, breast and lung cancers. The Int6 locus was initially identified as a common insertion site (CIS) in a genetic screen for transforming sequences in a breast cancer mouse model system. Insertion of mouse mammary tumor virus (MMTV) into this locus results in the production of an amino-terminal truncated gene product. Expression of the truncated Int6 product corresponds to cellular transformation in both in vivo and in vitro systems. This gene product plays a role in regulating translation initiation and is a component of the eukaryotic translation initiation factor 3 (eIF-3) complex. There is evidence that suggests that Int6 may impart a negative role in the general translational machinery while promoting an increase in the expression of a subset of stress-responsive genes. The eIF-3 complex is also required for disassembly and recycling of post-termination ribosomal complexes and subsequently prevents premature joining of the 40S and 60S ribosomal subunits prior to initiation. The eIF-3 complex specifically targets and initiates translation of a subset of mRNAs involved in cell proliferation, including cell cycling, differentiation and apoptosis, and uses different modes of RNA stem-loop binding to exert either translational activation or repression. Taken together, it is of great interest to further study the mechanism by which Int6 is involved in regulating cell growth. - Store vial at -20° C or below prior to opening. This vial contains a relatively low volume of reagent (25 µL). To minimize loss of volume dilute 1:10 by adding 225 µL of the buffer stated above directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect the volume at the bottom of the vial. Use this intermediate dilution when calculating final dilutions as recommended below. Store the vial at -20°C or below after dilution. Avoid cycles of freezing and thawing.
