Setdb2 is designed, produced, and validated as part of a collaboration between Rockland and the National Cancer Institute (NCI). Setdb2 and the neighboring gene Phf11 were linked to familial atopy/ asthma and are in a "critical region" commonly deleted in CLL. Setdb2 is a candidate histone methyltransferase as a member of the Su(var) 3-9 family of SET domain-containing enzymes. Other predicted domains in Setdb2 include a methylcytosine binding domain and a large bifurcation domain interrupting the putative histone methyltransferase (SET) domain whose function is unknown. Store vial at -20° C or below prior to opening. This vial contains a relatively low volume of reagent (25 µL). To minimize loss of volume dilute 1:10 by adding 225 µL of the buffer stated above directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect the volume at the bottom of the vial. Use this intermediate dilution when calculating final dilutions as recommended below. Store the vial at -20°C or below after dilution. Avoid cycles of freezing and thawing.
