       Caspase全称为含半胱氨酸的天冬氨酸蛋白水解酶(cysteinyl aspartate specific proteinase)。caspase是一类蛋白酶家族,成员较多,在人类,已经鉴定了11种不同的caspase,根据其蛋白酶序列的同源性可分为3个亚族:Caspase-1亚族包括 Caspase-1、4、5、11;Caspase-2亚族包括Caspase-2、9; Caspase-3亚族包括Caspase-3、6、7、8、10。起细胞凋亡执行者作用的是caspase3,6,7。

        【原理】FLICA试剂FAM-YVAD-FMK(Ex/Em = 488nm/530nm),是无细胞毒性的,细胞膜渗透抑制剂。进入每个细胞,并不可逆地与活化的caspase-1结合。任何未结合的FAM-YVAD-FMK-FLICA试剂扩散出细胞并被冲走。因此留在细胞内的绿色荧光直接体现了活性caspase-1酶活性的。荧光结果可通过荧光酶标仪、荧光显微镜、流式细胞仪进行分析。
