- Xanthine oxidase (XO, EC 1.17.3.2 ) is present in appreciable amounts in the liver and jejunumin healthy individuals. However, in various liver disorders, XO is released into circulation. Therefore, determination of serum XO level serves as a sensitive indicator of acute liver damage such as jaundice. This kit is an easy and sensitive assay to determine XO in variety of samples. In the assay, XO oxidizes xanthine to hydrogen peroxide (H2O2) which reacts stoichiometrically with OxiRed Probe to generate color (at lambda = 570nm) and fluorescence (at Ex/Em = 535/587nm). Since the color or fluorescence intensity is proportional to XO content, the XO activity can be accurately measured. The kit detects 1-100mU xanthine oxidase in 100ul reaction volume.Kit Components:X0980-28A: Assay Buffer, 1x25ml. Supplied as a liquid. Ready to use as supplied. X0980-28B: Probe, 1x200ul. Supplied as a liquid in DMSO. Ready to use as supplied. Warm to RT prior to use to completely melt frozen DMSO. Store at -20°C. Use within 2 months.X0980-28C: Enzyme Mix, 1x1vial. Supplied as a lyophilized powder. Dissolve with 220ul dH2O. Pipette to dissolve. X0980-28D: Substrate Mix, 1x1vial. Supplied as a lyophilized powder. Dissolve with 220ul dH2O. Pipette to dissolve.X0980-28E: Positive Control, 1x8ul. Dilute with 92ul ddH2O. Pipette to dissolve completely. Store at -20ºC. Use within 2 months.X0980-28F: H2O2 Standard (0.88M), 1x100ulStorage and Stability:Store the kit at -20°C. Stable for 6 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
