Ubiquitin propargylamide (Ub-PA) is a newly developed potent and specific deubiquitinating enzyme (DUB) inhibitor, N-terminally TAMRA labelled.
Acts as a DUB active site specific probe facilitating detection of DUBs in biological samples. Utility in the stabilisation of ubiquitin conjugates in cell lysates and tissue extracts and as a control inhibitor for the testing and characterisation of potential DUB activity modulators.
Ub-PA possess two unique capabilities:
1) Forms a covalent linkage with (the active site Cys residue of) a DUB that can be cleaved by acid treatment (5% aq. TFA), allowing for proteomic analyses
2) Targets all three major DUB families: UCH, USP and OTU
TAMRA-Ub-VME probe allows detection of DUB labeling by in-gel fluorescence. This direct and more sensitive read-out gives more distinct labelling paterns than can be achieved by immunoblotting. In addition, use of TAMRA-Ub-VME removes the potential for cross-reactivity of antibodies that can lead to background labeling.
