Ubiquitin C-terminal aminoluciferin derivative deubiquitinating enzyme (DUB) substrate. DUB cleavage of Ub-AML bond results in the release of aminoluciferin, which act as a substrate for luciferase allowing detection of luminescence as a read-out for DUB activity.
Ub-AML produces a stronger signal and has excellent signal to noise ratio compared to traditional fluorophores facilitating the rapid assessment of the activity of DUBs that do not readily process ubiquitin-AMC, whilst requiring much smaller amounts of DUB enzyme per assay. Potential utility in the high throughput screening of DUB activity modulators.
