CHIP is an ubiquitin E3 ligase enzyme associated with molecular chaperone mediated degradation of misfolded proteins.
CHIP binds both Hsp70 and Hsp90 and specifically catalyses ubiquitination of denatured but not native proteins in a chaperone-dependent manner. CHIP dependent K48-linked polyubiquitination of misfolded proteins that are resistant to refolding by molecular chaperones results in their degradation by the proteasome.
