Nrf2 is a transcription factor involved in oxidative stress response. Under normal or unstressed conditions, Nrf2 is sequestered in the cytoplasm by binding to Keap1, which acts as a substrate adaptor protein for its Cullin 3-based ubiquitination, resulting in the proteasomal degradation of Nrf2. Oxidative stress and / or increased levels of the autophagy receptor protein p62, which binds Keap1 directly, lead to a disruption of this process, allowing Nrf2 to translocate into the nucleus and subsequent induction of stress response genes. This association between Nrf2, KEAP1 and p62 suggests a role for selective autophagy in controlling the transcription of cellular defence enzyme genes and between oxidative stress and aggrephagy in which KEAP1 and p62 play a key role.
