NIX is a Bcl-2 related protein that is required for selective mitochondrial clearance by mitophagy in a ubiquitin independent manner during red blood cell differentiation in mammals. During this process unwanted mitochondria are targeted for degradation by the direct interaction of NIX, localised to the outer mitochondrial membrane, with LC3 / GABARAP proteins attached to the developing autophagosome membrane prompting their engulfment and subsequent destruction.
NIX has also been shown to play a more general role in mitophagy induction in response to oxidative stress and in the recruiting the ubiquitin E3 ligase Parkin to the mitochondria in ubiquitin mediated mitophagy.
