PINK1 is a mitochondrial kinase protein, mutated in autosomal recessive forms of Parkinson disease, that is normally processed in a membrane potential-dependent manner to maintain mitochondrial function, suppressing the need for autophagy. Upon mitochondrial depolarization, this processing is arrested so that PINK1 spans the outer mitochondrial membrane, recruiting the ubiquitin E3 ligase Parkin and subsequently p62 to facilitate ubiquitin mediated mitophagy.
