- The Stress-activated protein kinases (SAPK), also referred to as Jun N-terminal kinases (JNK), function in a protein kinase cascade transducing cellular stress signals (1–3). SAPK/JNKs are activated by a highly diverse group of extracellular signals, including: UV light, inflammatory cytokines, and a wide variety of cellular stresses (2–6). Activation occurs via phosphorylation at Thr183 and Tyr185 by the dual specificity enzyme SEK/MKK4 and as yet unidentified kinases. As dual phosphorylation of SAPK/JNK at Thr183/Tyr185 is essential for kinase activity, phosphorylation at this site is an excellent marker of SAPK/JNK activity (3–5). Activated SAPK in turn phosphorylates and activates several transcription factors including c-Jun at Ser63/73, and ATF-2 at Thr69/71. The SAPK/JNK (Thr183/Tyr185) Antibody Kit provides reagents and protocols for the rapid analysis of SAPK/JNK phosphorylation/activity. The Kit includes a phospho-antibody that recognizes SAPK/JNK only when phosphorylated at Thr183/Tyr185, a control SAPK/JNK antibody (phosphorylation-state independent), protein controls and protein markers for western blots, and our HRP Western Detection System.Applications: Suitable for use in Western Blotting, Immunocytochemistry. Other applications not tested.Recommended Dilution:Western blotting: 1:1000 1:1000Immunocytochemistry: 1:2000 1:200Optimal dilutions to be determined by the researcher.Storage and Stability:May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
