- Protein synthesis proceeds after formylation of methioninenby methionyl-tRNA formyl transferase (FMT) and transfer of thencharged initiator f-met tRNA to the ribosome. In eubacteria andneukaryotic organelles the product of this gene, peptide deformylasen(PDF), removes the formyl group from the initiating methionine ofnnascent peptides. In eubacteria, deformylation of nascent peptidesnis required for subsequent cleavage of initiating methionines bynmethionine aminopeptidase. The discovery that a natural inhibitornof PDF, actinonin, acts as an antimicrobial agent in some bacterianhas spurred intensive research into the design ofnbacterial-specific PDF inhibitors. In human cells, onlynmitochondrial proteins have N-formylation of initiatingnmethionines. Protein inhibitors of PDF or siRNAs of PDF block thengrowth of cancer cell lines but have no effect on normal cellngrowth. In humans, PDF function may therefore be restricted tonrapidly growing cells.
