- This gene encodes the class A macrophage scavengernreceptors, which include three different types (1, 2, 3) generatednby alternative splicing of this gene. These receptors or isoformsnare macrophage-specific trimeric integral membrane glycoproteinsnand have been implicated in many macrophage-associatednphysiological and pathological processes including atherosclerosis,nAlzheimer's disease, and host defense. The isoforms type 1 and typen2 are functional receptors and are able to mediate the endocytosisnof modified low density lipoproteins (LDLs). The isoform type 3ndoes not internalize modified LDL (acetyl-LDL) despite having thendomain shown to mediate this function in the types 1 and 2nisoforms. It has an altered intracellular processing and is trappednwithin the endoplasmic reticulum, making it unable to performnendocytosis. The isoform type 3 can inhibit the function ofnisoforms type 1 and type 2 when co-expressed, indicating a dominantnnegative effect and suggesting a mechanism for regulation ofnscavenger receptor activity in macrophages.
