- This gene is thought to play an important role in calciumnhomeostasis. The gene is expressed from two promoters and undergoesnextensive alternative splicing. The encoded set of proteins sharenvarying amounts of overlap near their N-termini but havensubstantial variations in their C-terminal domains resulting inndistinct functional properties. The longest isoforms (a and f)ninclude a C-terminal Aspartyl/Asparaginyl beta-hydroxylase domainnthat hydroxylates aspartic acid or asparagine residues in thenepidermal growth factor (EGF)-like domains of some proteins,nincluding protein C, coagulation factors VII, IX, and X, and thencomplement factors C1R and C1S. Other isoforms differ primarily innthe C-terminal sequence and lack the hydroxylase domain, and somenhave been localized to the endoplasmic and sarcoplasmic reticulum.nSome of these isoforms are found in complexes with calsequestrin,ntriadin, and the ryanodine receptor, and have been shown tonregulate calcium release from the sarcoplasmic reticulum. Somenisoforms have been implicated in metastasis.
