The protein encoded by this gene is a DNA-binding homeoboxnprotein and a transcriptional repressor. The encoded proteinnappears to repress transcription by recruiting histone deacetylasesnto TGF beta-responsive genes. This gene is amplified andnoverexpressed in some ovarian cancers, and mutations in this genencan cause holoprosencephaly.
