This gene encodes coagulation factor XII which circulatesnin blood as a zymogen. This single chain zymogen is converted to antwo-chain serine protease with an heavy chain (alpha-factor XIIa)nand a light chain. The heavy chain contains two fibronectin-typendomains, two epidermal growth factor (EGF)-like domains, a kringlendomain and a proline-rich domain, whereas the light chain containsnonly a catalytic domain. On activation, further cleavages takesnplace in the heavy chain, resulting in the production ofnbeta-factor XIIa light chain and the alpha-factor XIIa light chainnbecomes beta-factor XIIa heavy chain. Prekallikrein is cleaved bynfactor XII to form kallikrein, which then cleaves factor XII firstnto alpha-factor XIIa and then to beta-factor XIIa. The activenfactor XIIa participates in the initiation of blood coagulation,nfibrinolysis, and the generation of bradykinin and angiotensin. Itnactivates coagulation factors VII and XI. Defects in this gene donnot cause any clinical symptoms and the sole effect is thatnwhole-blood clotting time is prolonged.
