The cystatin superfamily encompasses proteins that containnmultiple cystatin-like sequences. Some of the members are activencysteine protease inhibitors, while others have lost or perhapsnnever acquired this inhibitory activity. There are three inhibitorynfamilies in the superfamily, including the type 1 cystatinsn(stefins), type 2 cystatins, and kininogens. This gene encodes anstefin that functions as a cysteine protease inhibitor, formingntight complexes with papain and the cathepsins B, H, and L. Thenprotein is one of the precursor proteins of cornified cell envelopenin keratinocytes and plays a role in epidermal development andnmaintenance. Stefins have been proposed as prognostic andndiagnostic tools for cancer.
