The calcium sensing receptor (CaSR) has been shown to play_x000D_ a major role in regulating parathyroid hormone secretion and_x000D_ subsequently influencing the calcium concentration of extracellular_x000D_ fluids. Based on sequence analysis of cDNA clones_x000D_ it appears that the CaSR is similar to the 7-transmembranedomain_x000D_ G-protein-coupled receptor superfamily. Changes in_x000D_ extracellular calcium are thought to modulate a balance between_x000D_ proliferation and differentiation in a variety of cell_x000D_ types. In normal primary keratinocytes and breast epithelial_x000D_ cells, proliferation is inhibited and elevated extracellular_x000D_ calcium levels trigger differentiation. Malignant transformations_x000D_ of these cell types are accompanied by a loss of responsiveness_x000D_ to the anti-proliferative effects of elevated extracellular_x000D_ calcium.
