- MEK1 and MEK2, also called MAP or ERK kinases, are dual specificity protein kinases that function in a mitogen activated protein kinase cascade controlling cell growth and differentiation (1–3). Activation of MEK1 and MEK2 occurs through phosphorylation of two serine residues at position 217 and 221, also in the L12 loop of subdomain VIII by Raf-like molecules. MEK1/2 is activated by a wide variety of growth factors, cytokines, and also by membrane depolarization and calcium influx (1–4). Constitutively active forms of MEK1/2 are sufficient for the transformation of NIH3T3 cells or the differentiation of PC-12 cells (4). MEK activates p44 and p42 MAP kinase by phosphorylating both threonine and tyrosine residues at sites located within the L12 loop of subdomain VIII of the catalytic domain. To date, p44/42 MAP kinases are the only known substrates for MEK1/2. Our phospho-MEK (Ser 217/221) antibody selectively recognizes active MEK, i.e., only when phosphorylated at Ser217/221 and hence is an excellent marker of MEK1/2 activity. The MEK1/2 (Ser217/221) Antibody Kit provides reagents and protocols for the rapid analysis of MEK1/2 (Ser217/221) phosphorylation status. The kit includes a phospho-antibody that recognizes MEK1/2 only when phosphorylated at Ser217/221, a control MEK1/2 antibody (phosphorylationstate independent), phosphorylated and nonphosphorylated MEK1/2 cell extracts for Western blotting and our HRP Western Detection System.Applications: Suitable for use in Western Blotting, Immunoprecipitation. Other applications not tested.Recommended Dilution:Western blotting: 1:1000Immunoprecipitation: 1:250Optimal dilutions to be determined by the researcher.Storage and Stability:May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
