- This kit includes all reagents required to measure MAP Kinase activity, including antibodies, substrate, buffers, and a chemiluminescent detection system. The use of two new phospho-antibodies, one to selectively precipitate active MAP Kinase and a second to detect MAPK-induced phosphorylation of Elk-1, results in a dramatic improvement over conventional IP/kinase assays. Signal to noise and overall sensitivity and specificity are improved, and best of all, radioactivity is totally eliminated from the assay. A monoclonal phospho-antibody to p44/42 MAP Kinase (Thr202 and Tyr204) is used to selectively immunoprecipitate active MAP Kinase from cell lysates. The resulting immunoprecipitate is then incubated with a Elk-1 fusion protein in the presence of ATP and Kinase Buffer; this allows immunoprecipitated active MAP Kinase to phosphorylate Elk-1 (1–3). Phosphorylation of Elk-1 at Ser383 is measured by western blotting using a phospho-Elk-1 (Ser383) antibody. Ser383 of Elk-1 is a major phosphorylation site by MAP Kinase and is required for Elk-1-dependent transcriptional activity (1–3). Selective analysis of Elk-1 phosphorylation at Ser383 results in improved specificity with high sensitivity and near zero background. When compared to 32P, this assay is actually more sensitive since the 32P label is highly diluted (usually to 50uM ATP), whereas the phosphoantibody can, at least in principle, detect every phosphorylated Elk-1 molecule. As a result, these changes dramatically improve signal to noise over conventional 32P assays and reduce autoradiography time to minutes instead of days.Applications: Suitable for use in Western Blotting, Immunoprecipitation. Other applications not tested.Recommended Dilution:Immunoprecipitation: 15ul of resuspended slurryWestern blotting: 1:1000Optimal dilutions to be determined by the researcher.Storage and Stability:May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C or colder. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
