Caspase-3 (3.4.22.56) is a member of the cysteine protease family of enzymes that cleaves at the C-terminal of an Aspartate residue. The cleavage event is a critical step in programmed cell death (including apoptosis, pyroptosis and necroptosis) and inflammation. Apoptosis is a tightly regulated event that plays a central role in the development and homeostasis of multi-cellular organisms. BioVision’s Caspase-3 Activity Assay Kit allows the detection and quantification of Caspase-3 activity by using a synthetic substrate Ac-DEVD-6-Amino-D-Luciferin. The substrate gets cleaved by Caspase-3 thereby generating luciferin which in turn is detected by the action of luciferase to generate highly sensitive signal proportional to the Caspase-3 activity. The assay is rapid and allows the high-throughput quantification of Caspase-3 Activity in cell lysates or adherent cells. It can be used to evaluate the onset of induction of apoptosis and/or evaluation of apoptosis inhibitors. The kit can detect Caspase-3 activity as low as 0.1 mU per well or detect as little as 40 apoptotic cells.
