- Multiplex ELISA kit for pro-inflammatory cytokines is designed for semi-quantitative and simultaneous determination of pro-inflammatory cytokines including interleukin 1alpha (IL-1alpha), interleukin 1 beta (IL-1beta), interleukin 6 (IL-6), interleukin 8 (IL-8), granulocyte macrophage colony stimulating factor (GM-CSF), monocyte chemotactic and activating factor (MCAF), and tumor necrosis factor alpha (TNF- alpha) in cell culture supernatant and other biological samples. This ELISA assay is a 3.5 hour solid phase immunoassay readily applicable to measure the levels of eight cytokines in cell culture supernatant, and other biological fluids. It showed no cross reactivity with various other proteins. This Multiplex ELISA is expected to be effectively used for investigations into the relationship inflammatory cytokine expression and various experimental, pathological and physiological conditions.The kit is intended FOR LABORATORY RESEARCH USE ONLY and is not for use in diagnostic or therapeutic procedures.Assay Time:3.5 hoursKit Components:I8432-08A: MICROTITER Strip, 1x96 wellsI8432-08B: Ab MIXTURE (BIOTIN), 1x6ml Mixture of Biotin conjugated anti-human cytokine antibodies.I8432-08C: Cytokines MIXTURE (HRP) CONJUGATE, 1x11ml, Mixture of Horseradish Peroxidase ConjugatesI8432-08D: STANDARD MIXTURE, 2x1vial, Two vials of lyophilized standard mixture. Each vial contains a buffered protein base and eight pro-inflammatory cytokines at different concentrations: IL-1alpha 1300pg, IL1beta 850pg, IL-6 1000pg, IL-8 2800pg, GM-CSF 1200pg, IFN-γ 2000pg, MCAF 1400pg, TNF- alpha 2800pg.I8432-08E: CALIBRATOR DILUENT I, 1x25ml, For serum/plasma testing. Newborn calf serum with PBS buffer and preservative. I8432-08F: CALIBRATOR DILUENT II, 1x25ml, For cell culture supernatant testing. Cell culture medium RPMI 1640 with newborn calf serum and preservative. I8432-08G: WASH BUFFER, 20X, 1x60ml, 20-fold concentrated solution of buffered surfactant.I8432-08H: SUBSTRATE A, 1x10ml, Buffered solution with H2O2I8432-08J: SUBSTRATE B, 1x10ml, Buffered solution with TMB.Storage and Stability:Store kit components at 4°C. Unopened kit components are stable for 6 months after receipt at 4°C. Reconstituted standards are stable for up to 30 days when stored sealed at 4°C.
