- The Mouse IgG2a ELISA kit is a highly sensitive two-site enzyme-linked immunoassay for measuring IgG2a in serum or plasma of mice.Principle of the Assay:In thiis assay the IgG2A present in samples reacts with the anti-IgG2a antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-IgG2a antibodies conjugated with horseradish peroxidase (HRP) are added. These enzyme-labeled antibodies form complexes with the previously bound IgG2a. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3’,5,5’-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of IgG2a in the sample tested; thus, the absorbance, at 450nm, is a measure of the concentration of IgG2a in the test sample. The quantity of IgG2a in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.Kit Components:I1904-90A1: Microtiter Plate, 1x96wells (12 x 8-well microwell strips in well holder frame). Each well is coated with affinity-purified anti-mouse IgG2a.I1904-90A2: IgG2a Calibrator, 1x1 vial. Mouse IgG2a calibrator at a concentration of 650ug/ml.I1904-90A3: IgG2a Antibody-HRP, 1x150ul. Affinity purified anti-mouse IgG2a antibody conjugated with horseradish peroxidase in a stabilizing buffer.I1904-90A4: Diluent Concentrate (5X), 1x50ml. 5X concentrated diluent running buffer.I1904-90A5: Wash Solution Concentrate (20X). 20X concentrated wash solution.I1904-90A6: TMB-Substrate Solution, 1x12ml. 3,3’,5,5’-tetramethybenzidine (TMB) and hydrogen peroxide in citric acid buffer at pH 3.3.I1904-90A7: Stop Solution, 1x12ml. 0.3M sulfuric acid.Materials Required But Not Provided:Precision pipette (2ul to 200ul) for making anddispensing dilutionsTest tubesMicrotitre washer/aspiratorDistilled or deionized H2OMicrotitre Plate readerAssorted glassware for the preparation of reagents and buffer solutionsTimerVortex mixerStorage and Stability:Kit stability is 6 months. Store kit components at 4°C except I1904-90A2 (IgG2a Calibrator). Upon receipt, aliquot I1904-90A2 and store at -20°C.
