- This Histone Methyltransferase Assay Reagent Kit is an accessory reagent kit for the analysis of histone methyltransferase activity.Quality Control:This kit is routinely evaluated by methylating core histones with P9004-01A (active PRMT1).Kit Components:H5203-85A: HMTase Assay Buffer (5X) 1x1ml. Supplied as a liquid containing 250mM Tris-HCl, pH 9.0, and 2.5mM dithiothreitol.H5203-85B: Core Histones: 1x200ug of precipitated chicken erythrocyte histones lyophilized from 200ul steriledistilled water. Reconstitute with sterile ddH2O to a concentration of 1mg/ml, aliquot to avoid repeated freezing and thawing and store the aliquots at -70°C.Core histones, including histone H1, were purified by acid extraction and TCA precipitation from chicken erythrocytes for use as a substrate in histone acetyl-transferase (HAT) assays. Purity ≥90% as judged by SDS PAGE and Coomassie blue staining of 10ug core histones.H5203-85C: P81 Phosphocellulose Squares: 1x200 pre-labeled squares. Strong cation exchanger of high capacity; ion exchange capacity is 18.0uEq/cm2. Thin (0.17mm) paper. Flow rate: 125mm/30 min. Very useful for the separation of biogenic amines, antibiotics, histamines, certain metals (Fe, Cu) at very low concentrations.Storage and Stability: Store H5203-85B (lyophilized powder) at -20°C; once reconstituted, store the liquid at -70°C. Store the other kit components at -20°C. Kit is stable for at least 6 months For maximum recovery of product, centrifuge the original vials after thawing and prior to removing the cap.
