- The Hsp90a BioAssay Kit is designed for identification of Hsp90a inhibitors using fluorescence polarization. The assay is based on the competition of fluorescently labeled geldanamycin for binding to purified recombinant Hsp90a. Hsp90a is a molecular chaperone with essential functions in maintaining transformation. Inhibition of Hsp90a functions has been shown to play a role in tumorigenesis and disease progression. The Hsp90a assay kit comes in a convenient 96-well format, with purified Hsp90a enzyme, fluorescently labeled geldanamycin and Hsp90a assay buffer for 100 enzyme reactions. The key to the Hsp90a BioAssay Kit is the fluorescently labeled geldanamycin. Using this kit, only one simple step on a microtiter plate is required for Hsp90a reactions. The fluorescently labeled geldanamycin is incubated with a sample containing Hsp90a enzyme to produce a change in fluorescent polarization that can then be measured using a fluorescence reader.Kit Components:H1832-86A: Hsp90a Recombinant Enzyme: 1x100ug. Store at -70°C.H1832-86B: Geldanamycin (FITC) 2.5uM: 1x30ul. Store at -70°C.H1832-86C: 5X Hsp90a Assay Buffer: 1x4ml. Store at -20°C.H1832-86D: Black, low binding NUNC black microtiter plate: 1x96 wells. Store at room temperature.APPLICATIONS: Suitable for studying enzyme kinetics and screening small molecular inhibitors for drug discovery and HTS applications. Other applications not tested.Storage and Stability:Store buffer at -20°C. Store enzyme and substrate at -70°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Avoid freeze/thaw cycles.
