- The Mouse Serum Amyloid A ELISA is a highly sensitive two-site enzyme-linked immunoassay (ELISA) for the quantitative determination of Serum Amyloid A (SAA) in mouse biological samples.Principle:In this assay the SAA present in sample reacts with the anti-SAA antibodies, which have been adsorbed to the surface of polystyrene microtiter wells. After the removal of unbound proteins by washing, anti-SAA antibodies conjugated with horseradish peroxidase (HRP), are added. This HRP-conjugated antibody forms a complex with the previously bound SAA. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3’,5,5’- tetramethylbenzidine (TMB). The quantity of bound enzyme is proportional to the concentration of SAA in the sample tested; thus, the absorbance, at 450 nm is a measure of the concentration of SAA in the test sample. The quantity of SAA in the test sample can be interpolated from the calibration curve constructed from the calibrators, and corrected for sample dilution.Kit Components:A2275-62A: Microtiter Plate, 12 x 8-well removable strips in well holder frame. Wells are coated with affinity-purified anti-mouse SAA.A2275-62B: Diluent Concentrate, 5X, 1x50ml. Concentrated diluent running buffer.A2275-62C: Wash Solution, 20X, 1x50mlA2275-62D: Antibody (HRP), 100X, 1x150ul. Concentrated affinity-purified anti-mouse SAA antibody conjugated with HRP in a stabilizing buffer.A2275-62E: TMB Substrate Solution, 1x12ml. Mixture of TMB and hydrogen peroxide in citric acid buffer, pH 3.3.A2275-62F: Stop Solution (0.3M H2SO4), 1x12ml. 0.3M Sulfuric acid. CAUTION: Avoid contact with skin.*A2275-62G: Mouse SAA Calibrator, 1x1 vial. Lyophilized mouse SAA.*A2275-62H: Positive Control, 1x50ul. 1:1000 Antigen-containing serum with 0.1% sodium azide. Concentration of mouse SAA is ~0.25mg/ml.Storage and Stability:Store *A2275-62G (Mouse SAA Calibrator) and A2275-62H (Positive Control) at -20°C. Store other components at 4°C. Stable for 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
