CXCL8 was originally discovered and purified as a neutrophil chemotactic and activating factor. It was also referred to as neutrophil chemotactic factor (NCF),neutrophil activating protein (NAP),monocytederived neutrophil chemotactic factor (MDNCF),T lymphocyte chemotactic factor (TCF),granulocyte chemotactic protein (GCP) and leukocyte adhesion inhibitor (LAI). Many cell types,including monocyte/macrophages,T cells,neutrophils,fibroblasts,endothelial cells,keratinocytes,hepatocytes,chondrocytes,and various tumor cell lines,can produce CXCL8 in response to a wide variety of proinflammatory stimuli such as exposure to IL-1,TNF,LPS,and viruses. An anti-h IL-8 monoclonal coating antibody is adsorbed onto microwells. IL-8 present in the sample or standard binds to antibodies adsorbed to the microwells. Following incubation unbound sample or standard are removed during a wash step. a Biotinylated anti-h IL-8 antibody is added and binds to IL-8 captured by the first antibody. Following incubation unbound Biotinylated anti-h IL-8 antibody is removed during a wash step. A Streptavidin-HRP is added and binds to Biotinylated anti-h IL-8 antibody. Following incubation unbound Streptavidin-HRP is removed during a wash step. A colored product is formed in proportion to the amount of IL-8 present in the sample. The reaction is terminated by addition of acid and absorbance is measured at 450nm.
