The interleukin 1 (IL-1) family of proteins consists of IL-1alpha,IL-1beta and the IL-1 receptor antagonist. IL-1alpha and IL-1beta bind to the same cell surface receptors and share biological functions. With the exception of skin keratinocytes, some epithelial cells, and certain cells of the central nervous system,IL-1 is not produced by unstimulated cells of healthy individuals. However,in response to stimuli such as those produced by inflammatory agents, infections,or microbial endotoxins,a dramatic increase in the production of IL-1 by macrophages and various other cell types is seen. IL-1 possesses a wide variety of biological activities and plays a central role in mediating immune and inflammatory responses. Although normal production of IL-1 is obviously critical for initiation of normal host responses to injury and infection, inappropriate or prolonged production of IL-1 has been implicated as playing a role in the production of a variety of pathological conditions including sepsis, rheumatoid arthritis, inflammatory bowel disease, acute and chronic myelogenous leukemia,insulin-dependent diabetes mellitus, and atherosclerosis.Sample Type:Cell culture supernatants, serum, plasma, and tissueIntended Use:The Rat IL-1beta BioAssay ELISA Kit is an enzyme-linked immunosorbent assay for the quantitative detection of Rat IL-1beta concentrations in cell culture supernatants, serum, plasma, and tissue.Sensitivity:15pg/mlRange:31.25-2000pg/mlSpecificity:Recognizes rat IL-1beta.Sample Volume:100ul/wellTest Principle:This assay employs the Sandwich immunoassay technique. An anti-r IL-12 p70 monoclonal coating antibody is adsorbed onto microwells. IL-12 p70 present in the sample or standard binds to antibodies adsorbed to the microwells. Following incubation unbound sample or standard are removed during a wash step. a Biotinylated anti-r IL-12 p70 antibody is added and binds to IL-12 p70 captured by the first antibody. Following incubation unbound Biotinylated anti-r IL-12 p70 antibody is removed during a wash step. A Streptavidin-HRP is added and binds to Biotinylated anti-r IL-12 p70 antibody. Following incubation unbound Streptavidin-HRP is removed during a wash step. A colored product is formed in proportion to the amount of IL-12 p70 present in the sample. The reaction is terminated by addition of acid and absorbance is measured at 450nm. A standard curve is prepared from seven IL-12 p70 standard dilutions and IL-12 p70 sample concentration determined.Kit Components:Aluminium pouches with a Microwell Plate: 8x6Standard: 1xVialBiotinylated antibody: 1xVialStreptavidin-HRP: 1xVialAssay Buffer (25ml/Vial): 1xVialWash Buffer Concentrate 20x (30ml/Vial): 1xVialTMB Substrate Solution: 1x6mlStop Solution (12ml/Vial): 1xVialPlate Covers- Adhesive strips: 2xstripsInstruction: 1xStorage and Stability:Store powder at 4°C liquid at -20°C.
