- IL1ra was originally isolated from the urine of patients with monocytic leukemia and has also been purified from adherent monocytes. The protein shows 26% amino acid homology to IL1b and 19% homology to IL1a. It will compete with either factor for receptor binding, but does not interact with either one. Human IL1ra will bind to both types of IL1 receptor (I and II) on human cells. In mouse, IL1 RII does not bind IL1ra. IL1ra has been shown to block the inflammatory responses induced by IL1 both in vitro and in vivo. Preclinical and clinical studies were done to test possible therapeutic applications for IL1ra in the treatment of sepsis, rheumatoid arthritis and chronic myelogenous leukemia.Sample Type:Serum and plasmaIntended Use:The kit is a sandwich enzyme immunoassay for the in vitro quantitative measurement of lL-1ra in mouse serum, plasma; and other biological fluids not being tested.Sensitivity:<1pg/mlRange:98.3-24,000pg/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of mouse IL-1ra.Precision:Intra CV<10%; Inter CV <15%Test Principle:This assay employs the fast quantitative sandwich enzyme immunoassay technique. Antibody specific for mouse IL-1ra has been pre-coated onto a microplate. Standards, samples and mouse IL-1ra mix are pipetted into the wells and any IL-1ra present is bound by the immobilized antibody. Following a wash to remove any unbound reagent, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-1ra bound in the initial step.
