- Interferon gamma (IFNgamma) exerts a wide range of immunoregulatory activities and is considered to be the prototype proinflammatory cytokine. Mature mouse IFNgamma exists as a noncovalently linked homodimer of 2025kD variably glycosylated subunits. It shares 86% amino acid sequence identity with rat IFNgamma and 38%-44% with human, porcine, and rhesus IFNgamma. IFNgamma is produced by a variety of immune cells under inflammatory conditions, notably by T cells and NK cells. It plays a key role in host defense by promoting the development and activation of Th1 cells, chemoattraction and activation of monocytes and macrophages, upregulation of antigen presentation molecules, and immunoglobulin class switching in B cells. It also exhibits antiviral, antiproliferative, and apoptotic effects.Sample Type:Serum and plasmaIntended Use:The kit is a sandwich enzyme immunoassay for the in vitro quantitative measurement of IFN-gamma in mouse serum, plasma; and other biological fluids not being tested.Sensitivity:4pg/mlRange:16.4-4000pg/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of mouse IFN-gamma.Precision:Intra CV<10%; Inter CV <15%Test Principle:This assay employs the fast quantitative sandwich enzyme immunoassay technique. Antibody specific for mouse IFN-gamma has been pre-coated onto a microplate. Standards, samples and mouse IFN-gamma mix are pipetted into the wells and any IFN-gamma present is bound by the immobilized antibody. Following a wash to remove any unbound reagent, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IFN-gamma bound in the initial step.
