- Sample Type:FecesIntended Use:This immunoassay kit allows for the in vitro semi-quantitative determination of mouse Rotavirus antigen concentrations in feces.Test Principle:The microtiter plate provided in this kit has been pre-coated with an antibody specific to Rotavirus antigen. Standards or samples are then added to the appropriate microtiter plate wells with a HRP-conjugated antibody preparation specific for Rotavirus antigen to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain Rotavirus antigen, HRP-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 2nm. The concentration of Rotavirus antigenin the samples is then determined by comparing the O.D. of the samples to the standard curve. Kit Components:Assay plate, 1x96 wellsPositive Control, 1x0.8mlNegative Control, 1x0.8mlHRP-conjugate, 1x6mlWash Buffer, 1x6mlSubstrate A, 1x5mlSubstrate B, 1x5mlStop Solution, 1x5mlStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
