- Progranulin (PGRN) also called epithelin precursor, proepithelin (PEPI), PC cell-derived growth factor (PCDGF), acrogranin, or paragranulin is a 593aa cysteine-rich protein of 68.5kD, that is typically secreted in a highly glycosylated 88kD form. The human Progranulin BioAssay ELISA kit is a sandwich Enzyme Linked-Immunosorbent Assay (ELISA) for quantitative determination of human progranulin in biological fluids.Sample Type:Serum, plasma, urine, cell culture supernatants and cerebrospinal fluid (CSF)Sensitivity:The lowest level of progranulin that can be detected by this assay is 32pg/ml. Range:0.063-4ng/ml Specificity:Recognizes natural and recombinant human Progranulin. It does not cross-react with human RBP4, human adiponectin, human Nampt, human leptin, human RELM-beta, human ANGPTL6, human FABP4, human TNF-alpha, human IL-33, human GPX3, human resistin, human clusterin, human vaspin, human PAl1, human ANGPTL3, mouse Nampt, rat Nampt.Test Principle:A polyclonal antibody specific for progranulin has been precoated onto the 96-well microtiter plate. Standards and samples are pipetted into the wells for binding to the coated antibody. After extensive washing to remove unbound compounds, progranulin is recognized by the addition of a biotinylated polyclonal antibody specific for progranulin (Detection Antibody). After removal of excess biotinylated antibody, HRP labeled streptavidin (Detector) is added. Following a final washing, peroxidase activity is quantified using the substrate 3,3’,5,5’-tetramethylbenzidine (TMB). The intensity of the color reaction is measured at 450nm after acidification and is directly proportional to the concentration of progranulin in the samples.Kit Components:Plate coated with human Progranulin antibody, 1x96wells10X Wash Buffer, 1x50mlDiluent 5X, 1x50mlDetection Antibody, 1x12mlDetector 100X (HRP Labeled Streptavidin), 1x150ulHuman Progranulin Standard, lyophilized, 1x8ngHuman progranulin QC sample, lyophilized, 1x200ulTMB Substrate solution, 1x12mlPlate Sealers, 3x plastic filmStop Solution 1x12mlStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
