Tetanolysin is one of a family of cholesterol binding toxins, known as cytolysins. These proteins form pores in the cytoplasmic membrane that allow ions and other molecules to pass through. Tetanolysin is purified from Clostridium tetani, and has a molecular weight of ~55kD. Cytolysins, including tetanolysin, generally have an LD50 of less than a microgram for mice, rabbits, and other laboratory animals. Their potent lethal and lytic properties are irreversibly inactivated by cholesterol and inhibited by oxidation. Conversely, the activity is somewhat enhanced by the addition of a reducing agent such as dithiothreitol. Tetanolysin, purified, is hemolytic to rabbit red blood cells at maximum dilutions of 10-5-10-6mg/ml. Tetanolysin is used to permeabilize cellular membranes. Thus, molecules that could not other- wise penetrate a membrane could be introduced to the endoplasm for the study of various cell functions. Tetanolysin has been used to introduce C3 exoenzyme in PC12 cells in order to inactivate Rho. Tetanolysin is reported to have a pore size of 20-50nm, similar to that of streptolysin O. Pores formed by streptolysin O have passed proteins as large as 50kD. Tetanolysin alters the ionic permeability of cells. For example, murine T cells have been permeabilized with tetanolysin so that the intracellular calcium level could be controlled; in this manner, the CD3 receptor’s ability to trigger inositol phospholipid hydrolysis has been examined at several calcium concentrations. Tetanolysin has been a useful tool for controlling intracellular ionic concentrations and introducing large molecules to the endoplasm. Source: Tetanolysin from Clostridium tetani Activity:Hemolytic activity of tetanolysin was determined with fresh rabbit red blood cells (RBC's). Since each cell type exhibits a different sensitivity, testing activity against a given cell type by using toxin at 0.1, 1.0, 10 and 100 HU/ml is highly recommended. Specific Activity:~118.34HU/ug Unit Definition:One hemolytic unit (HU) is the quantity of tetanolysin in a ml which causes 50% hemolysis of a rabbit RBC suspension at 37°C within 40 minutes. One HU is equivalent to 8.45ng of tetanolysin. Toxicity:Tetanolysin is a very potent toxin: microgram quantities are lethal for mice, rabbits and other laboratory animals. The lethal (cardiotoxic) and cytolytic properties are suppressed by oxidants. The biological properties are irreversibly lost in the presence of low concentrations of cholesterol and other 3-beta hydroxysterols which interfere with toxin binding to the target cells.Storage and Stability:Lyophilized powder may be stored at 4°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at 4°C. Reconstituted product is stable for 6 months at 4°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
