- The BioAssay ELISA Kit is a 1.5 hour solid-phase Competitive ELISA designed for the quantitative determination of Myosin-2 (MYH2) in rat serum, plasma, cell culture supernatants, tissue homogenates and body fluids. This kit for research use only, not for therapeutic or diagnostic applications.Detection Range:50-1000pg/mlSensitivity:1pg/mlPrecision:Intra-Assay CV: <10%Inter-Assay CV: <10%Specificity:This assay has high sensitivity and excellent specificity for detection of rat Myosin-2 (MYH2). No significant crossreactivity or interference with analogs was observed. Note: Crossreactivity detection between the target protein and all analogs has not been determined and crossreaction may exist in some cases.Assay Principle:Myosin-2 (MYH2) BioAssay ELISA Kit (Rat) utilizes the quantitative Competitive Enzyme Immunoassay technique, utilizing a monoclonal antibody and an conjugate to Myosin-2 (MYH2) (Rat). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Target Protein concentration since Protein from samples and Protein-HRP conjugate compete for the anti-Protein antibody binding site. Since the number of sites is limited, as more sites are occupied by Protein from the sample, fewer sites are left to bind the Protein-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Target Protein concentration in each sample is interpolated from this standard curve.Kit Components:197402A: Microtiter Strips, 1x48 wells or 1x96 wells.
