- The BioAssay ELISA Kit is a 1.5 hour solid-phase Competitive ELISA designed for the quantitative determination of mouse Glyceraldehyde 3 Phosphate Dehydrogenase (GAPDH) in serum, plasma, cell culture supernatants, body fluid and tissue homogenates. This kit for research use only, not forDetection Range:0.5-10ng/mlSensitivity:0.1ng/mlPrecision:Intra-Assay CV: 5.2, 7.9%Inter-Assay CV: 4.3, 5.4%Specificity:This assay has high sensitivity and excellent specificity for detection of GAPDH. No significant cross-reactivity or interference with analogs was observed. Note: Crossreactivity detection between GAPDH and all analogs has not been determined and crossreaction may exist in some cases.Assay Principle:Glyceraldehyde 3 Phosphate Dehydrogenase (GAPDH) BioAssay ELISA Kit (Mouse) utilizes the quantitative Competitive Enzyme Immunoassay technique, utilizing a monoclonal antibody and an conjugate to Glyceraldehyde 3 Phosphate Dehydrogenase (GAPDH) (Mouse). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Target Protein concentration since GAPDH from samples and GAPDH-HRP conjugate compete for the anti-GAPDH antibody binding site. Since the number of sites is limited, as more sites are occupied by GAPDH from the sample, fewer sites are left to bind the GAPDH-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards.
