- Intended Use:The BioAssay ELISA Kit is a 1.5 hour solid-phase competitive ELISA designed for the quantitative determination of Cross-linked N-Terminal Telopeptides of Type I Collagen (NTx-1) in rat serum, plasma, cell culture supernatant, tissue homogenates and body fluids. This kit is intended for research use only and may not be used for therapeutic or diagnostic applications.Detection Range:250-5000pg/mlSensitivity:1.0pg/mlPrecision:Intra-Assay %CV: 5.1, 8.2Inter-Assay %CV: 4.8, 5.9Specificity:This assay has high sensitivity and excellent specificity for detection of Cross-linked N-Terminal Telopeptides of Type I Collagen (NTx-1) (Rat). No significant crossreactivity or interference with analogs was observed. Note: Crossreactivity detection between the NTx-1 and all analogs has not been determined and crossreaction may exist in some cases.Assay Principle:Cross-linked N-Terminal Telopeptides of Type I Collagen (NTx-1) BioAssay ELISA Kit (Rat) utilizes the quantitative Competitive Enzyme Immunoassay technique, utilizing a monoclonal antibody and an conjugate to Cross-linked N-Terminal Telopeptides of Type I Collagen (NTx-1) (Rat). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NTx-1 concentration since NTx-1 from samples and NTx-1-HRP conjugate compete for the anti-NTx-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by NTx-1 from the sample, fewer sites are left to bind the NTx-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards.
