ImmunoTek Plus™ Microwell Peroxidase Substrate, Single ComponentMicrowell Peroxidase Substrate is a proprietary substrate formulation for peroxidase detection. The substrate is supplied as a one component liquid ready to use solution. Microwell Peroxidase Substrate has higher kinetic activity and can be used at additional wavelengths for even higher signal readings. Uniquely designed stop solutions for Microwell Peroxidase Substrate can even boost readings further. The substrate formulation is manufactured as a near colorless liquid with low background and a longer shelf life than most formulations. When Microwell Peroxidase Substrate is reacted with peroxidase, a soluble blue reaction develops that can be read at 405nm or 650nm. Manufactures two stop reagents tailored for Microwell Peroxidase Substrate. Depending on the stop reagent, the end point color will be stable for at least two hours. Product Use:Supplied as a one component ready to use formulation. The product should equilibrate to RT (25°C) before use. For immunoassay microwell applications, 100ul of substrate solution should be added to each well. A liquid soluble blue reaction develops. The reaction can be read at either 405nm or 650nm, with or without stopping. Microwell Peroxidase Substrate will produce signals when read at 405nm, the recommended wavelength, about 40% higher than conventional Microwell Peroxidase Substrate. Reading at 650nm will boost signal an additional 40% compared to 405nm readings. Optimally, absorbance values should be monitored before they exceed 2.0OD units. Two stop reagents tailored for Microwell Peroxidase Substrate: stops the reaction for end point readings at either 405nm or 650nm; and Turbo Stops the reaction for reading at 405nm with a signal boost of greater than twofold. Dilution of the substrate is not recommended and will alter its performance characteristics. To reduce reaction kinetics, it is recommended that analytes, antibodies or conjugates used in the assay be diluted. Applications:Not recommended for membrane or immunohistochemistry applications where a colored precipitation reaction is desired. Storage and Stability:Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
