产品描述: The Fluorogenic p300 BioAssay Kit is a homogeneous, fluorogenic assay designed to screen for inhibitors of p300 or to measure histone acetyltransferase (HAT) activity using cell extracts or purified enzymes. It comes in a convenient 96-well format, with all the reagents necessary for 100 fluorescent HAT activity measurements. No time-consuming washing steps are required. In addition, the kit includes purified p300 (also known as KAT3B) for use as a positive control. The Fluorogenic p300 Assay Kit is based on the transfer of an acetyl group from acetyl CoA to a peptide substrate. After incubation with acetyl CoA and the substrate, the P300 enzyme generates acetylated H3 peptide and CoASH. The thiol groups of CoASH can be detected with fluorogenic reagent at excitation = 360nm and emission = 460 nm.Components:P300 human recombinant enzyme, 1x10ugAcetyl CoA (1mM), 1x100ul10X HAT assay buffer, 1x5mlH3 peptide, 1x500ulFluorescence Developer, 1x250ulDeveloper Solution, 1x10mlHAT Stop Solution, 1x5mlblack, low binding NUNC black microtiter plate, 1 plateStorage and Stability:Store powder at 4°C liquid at -20°C.
