Pepsinogen II or pepsinogen C belongs to a group of aspartic proteinase zymogens (inactive form) that are activated in the acid pH of stomach to active enzyme pepsin. Pepsinogen II (PG II) is produced by chief cells and mucus neck cells of gastric mucosa, pyloric glands in the antrum and Brunner's glands in the proximal duodenum. Serum concentration of PG II reflects the histological status of gastric mucosa. The ratio of concentration of PGI to PGII in serum of normal subjects is about 4:1. The PGI/PGII ratio decreases linearly with increasing grade of atrophic gastritis in the corpus. The ratio is < 2.5 when the atrophic gastritis is advanced (moderate or severe) in the gastric corpus.SourceHuman Gastric MucosaApplications: Suitable for use in Western Blot. Other applications not tested.Recommended Dilution:Optimal dilutions to be determined by the researcher.Biological Activity:>1000Units/mg (After Activation) One unit will produce an OD280nm of 0.001/minute at pH 2.0 and 37°C, measured as TCA-soluble products using hemoglobin as substrate.Storage and Stability:Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
