- Granzyme B (GZMB, EC number 3.4.21.79) also known as Granzyme-2 is a serine protease stored in the granules of activated cytotoxic T cells and NK cells. Upon target cell contact, Granzyme B is directionally exocytosed and with the assistance of perforin enters the target cell. With its unique substrate specificity (Granzyme B prefers an aspartic acid residue at the P1 site of its substrates), Granzyme B processes and activates various pro-caspases thereby inducing apoptosis in the target cell. Intended Use:Measurement of Granzyme B activity in various biological samples or purified Granzyme B activity, Screening Granzyme B inhibitors, characterize/Study Granzyme B inhibitors.Test Principle:In the Granzyme B Activity BioAssay Kit, active human Granzyme B enzyme hydrolyzes the specific substrate to release the quench of fluorescent group, which can be detected fluorometrically at Ex/Em = 380/500nm. This BioAssay kit is rapid, reliable and sensitive and can detect Granzyme B activity in various biological samples.Kit Components:Granzyme B Assay Buffer, 1x25mlGranzyme B Substrate, 1x0.5mlPositive Control, Granzyme B Enzyme, human recombinant, 1x vialAFC Standard, 1mM, 1x0.1mlStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
