- Glutathione Reductase (GR, EC 1.8.1.7) catalyzes the NADPH-dependent reduction of oxidized glutathione (GSSG) to reduced glutathione (GSH), which plays an important role in the GSH redox cycle that maintains adequate levels of reduced GSH. A high GSH/GSSG ratio is essential for protection against oxidative stress. Sample Type:Cell and tissue culture supernatants, urine, plasma and serum, erythrocyte lysates, as well as many other biological fluids etc. Intended Use:The BioAssay can detect 0.1–40 mU/ml GR in various samples.
Test Principle:Glutathione Reductase BioAssay Kit is a highly sensitive, simple, direct and HTS-ready colorimetric assay for measuring GR activity in biological samples. In the assay, GR reduces GSSG to GSH, which reacts with 5, 5′- Dithiobis (2-nitrobenzoic acid) (DTNB) to generate TNB2- (yellow color, ymax = 405nm). Kit Components:GR Assay Buffer, 1x100ml3% H2O2, 1x1mlCatalase, lyophilized, 1x vialTNB Standard, 5mM, lyophilized, 1x vialDTNB, lyophilized, 1x vialNADPH, lyophilized, 2x vialGSSG, lyophilized, 1x vialGR Positive Control, 10mU, lyophilized, 1x vialStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
