- Anti-diphtheria toxoid antibodies are raised in response to vaccination with diphtheria toxoid protein. A patient’s response to the immunisation may be assessed, subsequently, by the serological determination of the anti-diphtheria toxoid antibody level using this quantitative enzyme immunoassay technique. It should be noted that the reference method, particularly in Europe, may be the Vero Cell Assay (VCA). This assay gives a measurement of the amountof ‘neutralising’ antibody of all immunoglobulin classes, not only IgG. Historically, comparison of commercial ELISA kits and the Vero Cell Assay has shown poor correlation, as the ELISA may detect a proportion of specific antibodies that may not neutralisethe diphtheria toxoid. As the assay utilizes the diphtheria vaccine for coating the plate, the assay gives a measurement of specific IgG antibodies produced in response to vaccination.Intended Use:BioAssay Kit is intended for the in-vitro measurement of specific IgG antibodies against diphtheria toxoid present in serum, in order to determine protective status pre- and post vaccination. Sufficient materials are supplied to allow a maximum of 39 samples to be tested in duplicate or 87 in single, with a seven point calibration curve and two controls. The seven point calibration curve can be reduced to five points if measurement below 0.037IU/ml is not required.Sensitivity: 0.006IU/mlRange: 0.004-3IU/ml Specificity: 96.60%Test Principle:Microwells are pre-coated with diphtheria toxoid antigen. The calibrators, controls and dilutedpatient samples are added to the wells and antibodies recognising the diphtheria toxoid antigen bind during the first incubation. After washing the wells to remove all unbound proteins, purified peroxidase labelled rabbit anti-human IgG (y) chain specific) conjugate is added. The conjugate binds to the captured human antibody and the excess unbound conjugate is removed by a further wash step. The bound conjugate is visualised with 3,3’,5,5 ’ tetramethylbenzidine (TMB) substrate which gives a blue reaction product, the intensity of which is proportional to the concentration of antibody in the sample. Phosphoric acid is added to each well to stop the reaction. This produces a yellow end point colour, which is read at 450nmKit Components:1. Diphtheria Toxoid Coated Wells, 12x8 well strips2. Stop Solution, 1x14mlStorage and Stability:Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
