Amprenavir is a first generation inhibitor of the HIV-1 aspartic protease (Ki = 0.04 nM) that blocks replication of HIV-1 in the human T cell MT-4 cell line (IC50 = 150 nM) as well as in peripheral blood lymphocytes (IC50 = 80 nM).{25257,25259,25258} Through this action, it blocks the post-translational processing of gag and gag-pol gene products into key structural proteins and replication enzymes.{25259} When used alone or in combination therapy with other antiretroviral agents, amprenavir decreases viral load and increases CD4+ cell counts.{25259}
