- Human PAI-1 ELISA Cytokine Development kit contains the key components required for the quantitative measurement of natural and/or recombinant Human PAI-1 in a sandwich ELISA format. Using the ELISA protocol described below, the recommended microplates, reagents and solutions, the components supplied in this kit are sufficient to assay Human PAI-1 in approximately 1000 ELISA plate wells.Detection Range:23–3000pg/mlCross-reactivity:When tested at 50ng/ml, the following human antigens did not exhibit significant cross-reactivity: Neuroserpin, Maspin, PAI-2, PEDF, and Vaspin.Kit Components:145960A: Capture Antibody, 1x50ug145960B: Detection Antibody, 1x100ug145960C: Human PAI-1 Standard, 1x1ug145960D: Avidin-HRP Conjugate, 1x60ulStorage and Stability:Store unopened kit at 4°C. Upon reconstitution, 145960A, 145960B & 145960C, are stable for 2 weeks at 4°C and for up to 6 months at -20°C. Upon receipt of the kit, 145960D should be aliquoted into 10 x 6ul vials and stored at ≤ -20°C; aliquots are stable for up to 12 months after receipt (avoid more than one freeze-thaw cycle).Reconstitution of Reagents:Capture Antibody: 50ug of antigen-affinity purified rabbit anti-Human PAI-1 containing 2.5mg D-mannitol. Centrifuge vial prior to opening. Reconstitute in 0.5ml sterile water for a concentration of 100ug/ml.Detection Antibody: 50ug of biotinylated antigen-affinity purified rabbit anti-Human PAI-1 containing 2.5mg D-mannitol. Centrifuge vial prior to opening. Reconstitute in 0.5ml sterile water for a concentration of 100ug/ml.Human PAI-1 Standard: 1ug of recombinant Human PAI-1 containing 2.2mg BSA, 11.0mg D-mannitol. Centrifuge vial prior to opening. Reconstitute in 1ml sterile water for a concentration of 1ug/ml.
