- Kallikrein-1, also known as tissue kallikrein, is a protein that in humans is encoded by the KLK1 gene. This serine protease generates Lys-bradykinin by specific proteolysis of kininogen-1. KLK1 is a member of the peptidase S1 family. Its gene is mapped to 19q13.3. In all, it has got 262-amino acids which contain a putative signal peptide, followed by a short activating peptide and the protease domain. The protein is mainly found in kidney, pancreas, and salivary gland, showing a unique pattern of tissue-specific expression relative to other members of the family. KLK1 is implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. Intended Use:KLK1 BioAssay ELISA Kit is a sandwich ELISA for quantitative detection of human KLK1 in cell culture supernates, cell lysates, serum and plasma (heparin, EDTA).Range:156pg/ml-10,000pg/mlSensitivity:<10 pg/mlSpecificity: Natural and recombinant human KLK1Crossreactivity:There is no detectable cross-reactivity with other relevant proteins.Kit Components:143943A: Microtiter Plate, 1x96 wells*143943B: KLK1 Standard, 2x1 vial143943C: Anti-human KLK1 (Biotin), 1x130ul (1:100 dilution)143696: Avidin-Biotin-Peroxidase Complex (ABC), 1x130ul (1:100 dilution)143697: Sample Diluent Buffer, 1x30ml143698: Antibody Diluent Buffer, 1x12ml143699: ABC Diluent Buffer, 1x12ml143700: TMB Color Developing Agent, 1x10ml143701: TMB Stop Solution, 2N H2SO4, 1x10mlStorage and Stability:Store *143943B powder at 4°C. Once reconstituted store at 4°C for up to 12 hours or at -20°C for up to 48 hours. Store other components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.Assay Summary:1. Add samples and standards and incubate the plate at 37°C for 90 minutes. Do not wash. 2. Add biotinylated antibodies and incubate the plate at 37°C for 60 minutes. Wash plate 3 times with 0.01M TBS or PBS.3. Add ABC working solution and incubate the plate at 37°C for 30 minutes. Wash plate 5 times with 0.01M TBS or PBS.4. Add TMB color developing agent and incubate the plate at 37°C in dark for 25-30 minutes.5. Add TMB stop solution and read.
