- Sandwich ELISA quantitative detection kit for Human IL-6 in cell culture supernates, serum and plasma (heparin, EDTA, citrate).The human interferon-beta 2 gene (IFNB2) product is identical to that for the B-cell stimulation factor-2 (BSF-2), the hybridoma growth factor (HGF) ("interleukin-6"), and the hepatocyte stimulating factor (HSF). Proteins derived from this gene mediate the plasma protein response to tissue injury (acute-phase response) and regulate the growth and differentiation of both B and T cells.1 Interleukin-6 (IL6) has come to be regarded as a potential osteoporotic factor because it has stimulatory effects on cells of the osteoclast lineage, and, thus, may play a role in the pathogenesis of bone loss associated with estrogen deficiency.2 IL-6 has many roles essential to the regulation of the immune response, hematopoiesis, and bone resorption.3 It is involved not only in the hepatic acute phase response but also in adipose tissue metabolism, lipoprotein lipase activity, and hepatic triglyceride secretion.4 Overproduction of IL-6, a proinflammatory cytokine, is associated with a spectrum of age-related conditions including cardiovascular disease, osteoporosis, arthritis, type 2 diabetes, certain cancers, periodontal disease, frailty, and functional decline.5 BSF-2 is a novel interleukin consisting of 184 amino acids.6 The standard product used in this kit is recombinant human IL-6, consisting of 184 amino acids with the molecular mass of 20.3kD.Assay Range:4.69pg/ml-300pg/mlSensitivity:<0.3 pg/mlCrossreactivity:There is no detectable cross-reactivity with other relevant proteins.Kit Components:143929A: Microtiter Strip 1x96wells pre-coated with anti-IL-6, Human143929B: Standard, Recombinant IL-6 Human, 10ng, 2x1 vial Lyophilized143929C: Anti-human IL-6 (Biotin), 1x130ulUSB Cat NoKit Component143696: Avidin-Biotin-Peroxidase Complex (ABC), 1x130ul (1:100 dilution)143697: Sample diluent buffer, 1x30ml143698: Antibody diluent buffer, 1x12ml143699: ABC diluent buffer, 1x12ml143700: TMB color developing agent, 1x10ml143701: TMB stop solution, 2N H2SO4, 1x10mlStorage and Stability:Store *143929B powder at 4°C. Once reconstituted store at 4°C for up to 12 hours or at -20°C for up to 48 hours. Store other components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.Assay Summary:1. Add 100ul of samples and standards and incubate the plate at 37°C for 90 min. Do not wash.2. Add 100ul biotinylated antibody and incubate the plate at 37°C for 60 min. Wash plate 3 times with 0.01M TBS or PBS.3. Add ABC working solution and incubate the plate at 37°C for 30min. Wash plate 5 times with 0.01M TBS or PBS.4. Add TMB color developing agent and incubate the plate at 37°C in dark for 20-25 min.5. Add 100ul TMB stop solution and read.
