- Interleukin-22 (IL-22), also known as ILTIF, is protein that in humans is encoded by the IL22 gene. IL-22 a member of a group of cytokines called the IL-10 family or IL-10 superfamily a class of potent mediators of cellular inflammatory responses. Using FISH, the IL22 gene is mapped to chromosome 12q15, close to the IFNG and the herpesvirus saimiri-induced AK155 genes. IL-22 can contribute to immune disease through the stimulation of inflammatory responses, S100s and defensins. IL-22 also promotes hepatocyte survival in the liver and epithelial cells in the lung and gut similar to IL-10. In some contexts, the pro-inflammatory versus tissue-protective functions of IL-22 are regulated by the often co-expressed cytokine IL-17A.Intended Use:For quantitative detection of human IL-22 in cell culture supernates, serum and plasma (heparin, EDTA).Assay Range:15.6pg/ml-1000pg/mlSensitivity:<10 pg/mlSpecificity:Natural and recombinant human IL-22Kit Components:143911A: Microtiter Strip, 1x96 wells*143911B: Standard, 10ng, 2x1vial143911C: Anti-human IL-22 (Biotin), 130ulUSB Cat NoKit Component143696: Avidin-Biotin-Peroxidase Complex (ABC), 1x130ul (1:100 dilution)143697: Sample Diluent Buffer, 1x30ml143698: Antibody Diluent Buffer, 1x12ml143699: ABC Diluent Buffer, 1x12ml143700: TMB Color Developing Agent, 1x10ml143701: TMB Stop Solution, 2N H2SO4, 1x10mlStorage and Stability:Store *143911B powder at 4°C. Once reconstituted store at 4°C for up to 12 hours or at -20°C for up to 48 hours. Store other components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.Assay Summary:1. Add 100ul of samples and standards and incubate the plate at 37°C for 90 minutes. Do not wash.2. Add 100ul biotinylated antibody and incubate the plate at 37°C for 60 minutes. Wash plate 3 times with 0.01M TBS or PBS.3. Add 100ul of ABC working solution and incubate the plate at 37°C for 30 minutes. Wash plate 5 times with 0.01M TBS or PBS.4. Add 90ul of TMB color developing agent and incubate the plate at 37°C in dark for 20-25 minutes.5. Add 100ul TMB Stop Solution and read.
