- Glutathione peroxidase 3 (GPX3; GSHPX3) is a selenium-dependent extracellular enzyme, protecting cells and enzymes from oxidative damage by catalyzing the reduction of hydrogen peroxide, lipid peroxides and organic hydroperoxides. GPX3 is involved in inflammatory bowel disease, in asthma and in diabetes. The activity of GPX3 is significantly reduced in the plasma and tissue of cancer patients. Recently, GPX3 was suggested to serve as a molecular marker for the diagnosis of clear cell type ovarian adenocarcinoma.The Glutathione Peroxidase 3 (GPX3) BioAssay ELISA Kit is to be used for the in vitro quantitative determination of human GPX3 in serum, plasma and cell culture supernatant. This ELISA Kit is for research use only.Assay Principle:This assay is a sandwich Enzyme Linked-Immunosorbent Assay (ELISA) for quantitative determination of human GPX3 in biological fluids. A polyclonal antibody specific for GPX3 has been precoated onto the 96-well microtiter plate. Standards and samples are pipetted into the wells for binding to the coated antibody. After extensive washing to remove unbound compounds, GPX3 is recognized by the addition of a biotinylated polyclonal antibody specific for GPX3 (Detection Antibody). After removal of excess biotinylated antibody, HRP labeled streptavidin (Detector) is added. Following a final washing, peroxidase activity is quantified using the substrate 3,3’,5,5’-tetramethylbenzidine (TMB). The intensity of the color reaction is measured at 450 nm after acidification and is directly proportional to the concentration of GPX3 in the samples.Kit Components:044865A: Microtiter Strips, 1x96wells (6x16 wells)044865B: Wash Buffer, 10X, 2x30ml044865C: ELISA Buffer, 10X, 2x30ml044865D: Detection Antibody, 1x20ul044865E: Streptavidin (HRP) (Lyophilized), 1x2ug*044865F: GPX3 Standard (Lyophilized), 1x1vial044865G: TMB Substrate Solution, 1x12ml044865H: Stop Solution, 1N Sulfuric Acid, 1x12mlStorage and Stability:Store *044865F powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
