- DNER is an activator of the Notch1 pathway. DNER is expressed in several types of neurons, including cortical and hippocampal pyramidal neurons, cerebellar granule cells, and Purkinje cells. DNER has been shown to be an antigen recognized by anti-Tr, one of the best described autoantibodies in paraneoplastic cerebellar degeneration (PCD) combined with Hodgkin lymphoma (HL). Presence of anti-Tr antibodies in patients with PCD and HL (or HL only) could now be screened quickly and reliably. DNER also contributes to the morphological and functional maturation of Bergmann glia via the Notch signaling pathway, and is essential for precise cerebellar development. DNER might play a role in the pathophysiology of joint destruction in osteoarthritis. DNER is expressed in early pancreatic progenitors and has been implicated in adipogenesis by acting as an anti-adipogenic factor, suggesting that a metabolic regulatory role is associated with this Notch ligand molecule.
