SREBF1 (Sterol Regulatory Element Binding Transcription Factor 1) is a transcriptional activator required for lipid homeostasis. It regulates transcription of the LDL receptor gene as well as the fatty acid and to a lesser degree the cholesterol synthesis pathway. It binds to the sterol regulatory element 1 (SRE-1) (5-ATCACCCCAC-3), which is a motif that is found in the promoter of the low-density lipoprotein receptor gene and other genes involved in sterol biosynthesis. SREBP1 has dual sequence specificity binding to both an E-box motif (5-ATCACGTGA-3) and to SRE-1 (5-ATCACCCCAC-3). Store vial at -20° C or below prior to opening. This vial contains a relatively low volume of reagent (25 µL). To minimize loss of volume dilute 1:10 by adding 225 µL of the buffer stated above directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect the volume at the bottom of the vial. Use this intermediate dilution when calculating final dilutions as recommended below. Store the vial at -20°C or below after dilution. Avoid cycles of freezing and thawing.
